Diego began his career with Botafogo de Ribeirão Preto in 2001 and quickly excelled in several tournaments, winning the best goalkeeper award in all of them. At 19, he transferred to Atlético Mineiro and was the team's standout performer in their return to the Brasileirão Série A in 2006. The following year, he won the Campeonato Mineiro, again being named best goalkeeper.

That same year, he was signed by Almería in Spain and went 618 minutes without conceding a goal and saved seven penalties out of 13 taken in the league. In 2011, he joined Valencia and maintained his reputation as a penalty saver.
